Ontdooien van PRF-cellijnen<\/strong><\/h4>\n

  1. Ontdooi de cellen snel in een waterbad van 37\u02daC.<\/li>\n
  2. Veeg de buitenkant van het flesje schoon met 70%-ethanol.<\/li>\n
  3. Breng de ontdooide cellen over in een T25-fles met 5 ml groeimedium.<\/li>\n
  4. Plaats de kolf een nacht in een broedstoof bij 37\u02daC.<\/li>\n
  5. Bekijk de volgende dag onder de microscoop of de cellen zich hebben gehecht.<\/li>\n
  6. Verwijder de media en vervang ze door nieuwe groeimedia.<\/li>\n<\/ol>\n<\/div>\n<\/div>\n<\/div>\n<\/div>\n<\/div>\n<\/div>\n

    Subculturen van PRF-cellijnen<\/strong><\/h4>\n
      \n
    1. Cellen zouden gesplitst moeten worden als ze samenvloeien.<\/li>\n
    2. Om cellen te splitsen (de opgegeven volumes zijn gebaseerd op de veronderstelling dat de cellen zich in een T25 bevinden):
      a) Verwijder de media met behulp van een steriele techniek en spoel de kolf met 2-3 ml steriele HBSS. Verwijder de HBSS en gooi deze weg.
      b) Voeg 1 ml trypsine toe en incubeer 2-3 minuten. Cellen moeten onder een omgekeerde microscoop worden gecontroleerd om te bepalen of ze zijn begonnen met ronddraaien, optillen en drijven. Als de cellen na 3 minuten niet loskomen, kunt u nog eens 1-2 minuten incuberen.
      c) Draai de dop vast en kantel de kolf voorzichtig van links naar rechts om alle cellen los te maken. De kolf kan indien nodig lichtjes op de zijkant worden getikt om de cellen los te maken.
      d) Voeg 4 ml nieuwe media toe aan de kolf zodra de cellen drijven om de trypsine te inactiveren. Spoel de zijkanten van de kolf meerdere keren af met de nieuwe media om alle cellen van het plastic en in de oplossing te wassen. Verwijder alle vloeistof met cellen en giet over in een conische 15 ml (of 50 ml als u 3 of meer kolven samenvoegt).
      e) Neem met behulp van een steriele techniek een aliquot om te tellen op een hemocytometer.
      f) Terwijl u de cellen telt, moet de rest van de oplossing gedurende 5 minuten bij 1000 toeren per minuut in de klinische centrifuge worden gecentrifugeerd.<\/li>\n
    3. Nadat u uw celtelling hebt berekend, plaatst u de cellen op 2,5 x 105<\/sup>\u00a0<\/span>cellen per T25-filtertopkolf.<\/li>\n
    4. De cellen moeten elke 2-3 dagen gevoed worden met vers groeimedium.<\/li>\n<\/ol>\n

      Bevriezen:
      <\/u><\/strong><\/h4>\n

      Cellen moeten worden ingevroren bij minimaal 5 x 105<\/sup>\u00a0<\/span>cellen\/ml\/cryovial in groeimedia met 10% DMSO en 30% FBS en vervolgens 's nachts in een isopropanolvrieskamer bij -80\u02daC geplaatst. De volgende dag overbrengen naar vloeibare stikstof.<\/p>\n

      1. Thaw cells rapidly in a 37\u02daC water bath.<\/li>\r\n \t
      2. Wipe the outside of the vial with 70% ethanol.<\/li>\r\n \t
      3. Transfer the thawed cells to a T25 flask containing 5 ml of growth media.<\/li>\r\n \t
      4. Place flask in 37\u02daC incubator overnight.<\/li>\r\n \t
      5. The following day, observe under the microscope to ensure cells have attached.<\/li>\r\n \t
      6. Remove media and replace with fresh growth media.<\/li>\r\n<\/ol>\r\n[\/vc_column_text][\/vc_column][\/vc_row][vc_row][vc_column][vc_column_text]Subculturing PRF cell lines<\/u><\/strong>\r\n
          \r\n \t
        1. Cells should be split when confluent.<\/li>\r\n \t
        2. To split cells (volumes given are assuming cells are in a T25):\r\na)\u00a0 Using sterile technique, remove media and rinse flask with 2-3 ml of sterile HBSS. Remove and discard HBSS.\r\nb)\u00a0 Add 1 ml trypsin and incubate for 2-3 minutes. Cells should be checked under an inverted microscope to determine if they have begun to round up, lift and float.\u00a0 If cells are not detached after 3 minutes, you may incubate another 1-2 minutes.\r\nc)\u00a0 Tighten cap and gently tip flask from side to side to dislodge all cells. Flask may be tapped lightly on side to loosen cells if needed.\r\nd)\u00a0 Add 4 ml of new media to flask as soon as cells are floating to inactivate the trypsin. Rinse down sides of flask several times with the new media to wash all cells off the plastic and into the solution. Remove all liquid containing cells and transfer to a 15ml conical (or 50 ml if you are pooling 3 or more flasks).\r\ne)\u00a0 Using sterile technique, remove an aliquot for counting on a hemocytometer.\r\nf)\u00a0 While you are counting cells, the rest of the solution should be spun in the clinical centrifuge for 5 minutes at 1000 rpm.<\/li>\r\n \t
        3. After calculating your cell count, plate cells at 2.5 x 105<\/sup> cells per T25 filter top flask.<\/li>\r\n \t
        4.
