Progeria is caused by a single typo in a child’s DNA. Out of the roughly 3 billion letters in the human genome, just one is wrong – a T where a C should be -- in a gene called LMNA. That single misspelling causes the body to produce a toxic protein called progerin, which accumulates in cells, causes rapid aging, and ultimately destroys the cardiovascular system.

For most of medical history, there was no way to correct a mutation at the genetic level. That has changed.

Base-editing: a precise molecular rewrite

Base editing is a precise, next-generation form of gene editing. Unlike earlier gene-editing tools, which cut the DNA strand and risked unintended errors, a base editor acts like a molecular pencil, changing a single letter in the genetic code without breaking the DNA double helix. In this case, SamPro-2 is a base editor that was developed specifically for Progeria. It finds the wrong T in the LMNA gene and converts it back to the correct C.

The result: cells stop making progerin. The disease loses its engine.

SamPro-2 uses a delivery vehicle called AAV9, which acts like a molecular FedEx truck. It is a modified viral shell that carries the gene editor directly into a patient’s cells. AAV9 does not replicate, does not cause disease, and is the delivery component of the FDA-approved therapy, Zolgensma, for Spinal Muscular Atrophy. AAV9 is currently being studied in a variety of clinical trials for other diseases. For Progeria, this delivery truck was designed to be delivered to cells intravenously, meaning it can travel through the bloodstream to reach the cardiovascular system and other organs throughout the body.

This systemic reach is essential because Progeria is a multisystem disease.

SamPro-2 is the PATH to the Cure, Not THE Cure

Today’s gene-editing technologies cannot yet reach 100% of cells in the human body. SamPro-2 is designed to correct a substantial portion, especially in the cardiovascular system where Progeria causes its most life-threatening damage. This has the potential to dramatically slow disease progression, extend lifespan, and improve quality of life. As we learn from each clinical trial, PRF is committed to developing next-generation versions of SamPro-2 toward, ultimately, a complete cure.

Thousands of diseases are caused by a single misspelled DNA letter.
